The endoplasmic reticulum (ER) extends throughout a cell and plays a critical role in maintaining cellular homeostasis.
Changes in ER shape could provide a clue to explore the mechanisms that underlie the fate determination of neurons after
axon injury because the ER drastically changes its morphology under neuronal stress to maintain cellular homeostasis and
recover from damage. Because of their tiny structures and richness in the soma, the detailed morphology of the ER and... Read more

Characterization of the bone marrow adipocyte niche with three-dimensional electron microscopy
Unlike white and brown adipose tissues, the bone marrow adipocyte (BMA) exists in a microenvironment containing unique populations of hematopoietic and skeletal cells.
To study this microenvironment at the subcellular level, we performed a three-dimensional analysis of the ultrastructure of the BMA niche with focused ion beam scanning electron microscopy (FIB-SEM). This revealed that BMAs display hallmarks of metabolically active cells including polarized lipid deposits, a dense mitoch... Read more

Loss of adult skeletal muscle stem cells drives age-related neuromuscular junction degeneration
Neuromuscular junction degeneration is a prominent aspect of sarcopenia, the age-associated loss of skeletal muscle integrity. Previously, we showed that muscle stem cells activate and contribute to mouse neuromuscular junction regeneration in response to denervation (Liu et al., 2015). Here, we examined gene expression profiles and neuromuscular junction integrity in aged mouse muscles, and unexpectedly found limited denervation despite a high level of degenerated neuromuscular junctions. In... Read more

Mitochondria-rough-ER contacts in the liver regulate systemic lipid homeostasis
In this work, we studied mitochondria-rER contacts in vivo by serial section electron tomography (SSET) and 3D reconstruction analysis of cryo-fixed mouse tissue samples. We characterized this inter-organelle association as mitochondria tightly wrapped by sheets of curved rER (wrappER). Further, we used multi-omics and genetic approaches to obtain evidence that the wrappER is a distinct intracellular compartment and demonstrate the importance of wrappER-mitochondria contacts for v... The high strength at moderate weight in combination with superior corrosion and fatigue properties makes carbon-fiber-reinforced polymer (CFRP) an attractive material for lightweight applications in aerospace. Nonetheless, besides several benefits, CFRP components also bear significant risks like a low resistance to impact damage. […] In this work, we present a multimodal approach to three-dimensionally quantify and visualize fiber orientation and resin-rich areas in carbon-fiber-reinf... Read more

Additive manufacturing (AM) of aluminum alloy components has drawn broad attention from industrial customers in recent years. With the advantages of low cost and high deposition rate, wire + arc additive manufacturing (WAAM) has been recognized as a promising AM technology in producing large-scale aluminum alloy structural parts. However, the adverse effect of internal defects on mechanical properties has limited the application of WAAM aluminum alloys. Micropores, as one of the most harmfu... Read more

Cognition based bTBI mechanistic criteria; a tool for preventive and therapeutic innovations
Blast-induced traumatic brain injury has been associated with neurodegenerative and neuropsychiatric disorders. To date, although damage due to oxidative stress appears to be important, the specific mechanistic causes of such disorders remain elusive. Here, to determine the mechanical variables governing the tissue damage eventually cascading into cognitive deficits, we performed a study on the mechanics of rat brain under blast conditions. To this end, experiments were carried out to analyse... Read more

Electron Beam Melting (EBM) is an Additive Manufacturing (AM) process which can be used to fabricate complex 3D metal components. Ti-6Al-4V is one of the high value aerospace materials that has been widely used in the EBM process. However, defects such as gas pores and lack of fusion are often found in the as-built components, which can make a significant difference to the fatigue resistance of the parts. The Manufacturing Technology Centre (MTC) uses X-ray Computed Tomography (XCT) and 3D im... Read more

For many years, SFRPs (short fibre reinforced polyamides) have been used in the automotive industry as a means to reduce vehicle weight. However, their complex anisotropic and heterogeneous microstructure requires sophisticated material characterisation and simulation. This study aims at presenting the simulation strategy adopted by an automotive company address these challenges. The manufacturing process is first simulated and correlated with tomography analysis. Then, based on the numerical... Read more

3D virtual reconstruction of the Kebara 2 Neandertal thorax
The size and shape of the Neandertal thorax has been debated since the first discovery of Neandertal ribs more than 150 years ago, with workers proposing different interpretations ranging from a Neandertal thoracic morphology that is indistinguishable from modern humans, to one that was significantly different from them. Here, we provide a virtual 3D reconstruction of the thorax of the adult male Kebara 2 Neandertal. Our analyses reveal that the Kebara 2 thorax is significantly different but ... Read more

Understanding the near-threshold crack growth behavior in an aluminum alloy by x-ray tomography
The near-threshold behavior of long cracks is evaluated using pre-cracked flat dogbone specimens of a commercial aluminum alloy in two heat treatment states. Once the threshold was known, the crack initially introduced by cyclic compression was propagated under load control with approximately
constant range of the stress intensity factor at values close to the threshold values. The 3D morphology of the crack and the spatial distribution of the primary precipitates obtained by µ-CT were... Read more

3D characterization of the fracture mechanisms of a Fe-rich Al-Si-Cu alloy
The effect of the defect size and morphology on the fatigue damage evolution was analysed in a recycled Al-Si-Cu alloy by micro-computed tomography and scanning electron microscopy. Fatigue tests were performed and the different crack initiation scenarios were characterized and classified. The interaction between shrinkage and gas pores was the key crack initiation mechanism and the ß-Al5FeSi particles did not play any role in the crack initiation phase. However, crack path analysis indicate... Read more

Three dimensional (3D) composites were proposed over 40 years ago in an attempt to overcome the shortcomings of 2D laminates, by incorporating fibres into the through-thickness direction. 3D weaving offer significant manufacturing benefits as well as creating versatile textiles having a range of 3D architectures.
The development of fatigue damage in a glass fibre modified layer-to-layer three dimensional (3D) woven composite has been followed by time-lapse X-ray computed tomograp... Read more

An eosauropterygian skeleton found in the Middle Triassic (upper Anisian) Gutenstein Formation of the Fatric Unit (Demänovská dolina Valley, Low Tatra Mountains, Slovakia) represents the earliest known occurrence of marine tetrapods in the Western Carpathians. The specimen represents a partly articulated portion of the postcranial skeleton (nine dorsal vertebrae, coracoid, ribs, gastral ribs, pelvic girdle, femur and one zeugopodial element). It is assigned to the Pachypleurosauria, more pr... Read more
