In recent years new methodologies and workflow pipelines for acquiring correlated fluorescence microscopy and volume electron microscopy datasets have been extensively described and made accessible to users of different levels. Post-acquisition image processing, and particularly correlation of the optical and electron data in a single integrated three-dimensional framework can be key for extracting valuable information, especially when imaging large sample volumes such as whole cells or tissu... X-ray computed tomography (XCT) has been shown to reveal the true extent of ductile damage below the fracture surface of failed test specimens, which is often significantly underestimated when probed using 2D serial sectioning techniques and a microscope, since a single plane of material may only exhibit only a handful of resolvable voids.
In contrast XCT offers the capability to generate large datasets consisting of hundreds, if not thousands, of individually resolvable voids, where e... Read more

Cortical bone is permeated by a system of pores, occupied by the blood supply and osteocytes. With ageing, bone mass reduction and disruption of the microstructure are associated with reduced vascular supply. Insight into the regulation of the blood supply to the bone could enhance the understanding of bone strength determinants and fracture healing. Using synchrotron radiation-based computed tomography, the distribution of vascular canals and osteocyte lacunae was assessed in murine cortica... Read more

Optical Resolution Photoacoustic Microscopy of Ovary and Fallopian Tube
Ovarian cancer is the leading cause of death among gynecological cancers, but is poorly amenable to preoperative diagnosis. In this study, we investigate the feasibility of “optical biopsy,” using high-optical-resolution photoacoustic microscopy (OR-PAM) to quantify the microvasculature of ovarian and fallopian tube tissue. The technique is demonstrated using excised human ovary and fallopian tube specimens imaged immediately after surgery.
This report describes the first applicatio... Read more

3D Dissection of Structural Membrane-Wall Contacts in Filamentous Moss Protonemata
Cell-to-cell contact is essential for communication and development of multicellular organisms. A prerequisite is the passage through membranes. That way, molecular exchange and information flow is regulated via hormones, membrane proteins and pores.
In plants, the rigid cell walls prevent large membrane contact areas between protoplasts. Only plasmodesmata, minute channels between adjacent cells, form direct connections. Often, molecular data of the proteins involved are manifold but t... Read more

Each pulmonary segment is an anatomical and functional unit. However, it is fundamentally difficult to precisely distinguish every pulmonary segment using the conventional pulmonary intersegmental planes from computed tomography images. Building arteriopulmonary segments is likely to be an effective way to identify pulmonary segments.
The three-dimensional reconstructed images showed the branches of the pulmonary artery ramified up to their eighth order covering the entire lung as well... Read more

For decades, clearing and staining with Alcian Blue and Alizarin Red has been the gold standard to image vertebrate skeletal development. Here, we present an alternate approach to visualise bone and cartilage based on X-ray microCT imaging, which allows the collection of genuine 3D data of the entire developing skeleton at micron resolution.
Our novel protocol is based on ethanol fixation and staining with Ruthenium Red, and efficiently contrasts cartilage matrix, as demonstrated in wh... Read more

Progressive transformation of the otic placode into the functional inner ear during gestational development in humans leads to the acquisition of hearing perception via the cochlea and balance and spatial orientation via the vestibular organ.
Using a correlative approach involving micro-computerized tomography (micro-CT), transmission electron microscopy and histological techniques we were able to examine both the morphological and cellular changes associated with human inner ear devel... Read more

In biomedical research, a huge variety of different techniques is currently available for the structural examination of small specimens, including conventional light microscopy (LM), transmission electron microscopy (TEM), confocal laser scanning microscopy (CLSM), microscopic X-ray computed tomography (microCT), and many others. Since every imaging method is physically limited by certain parameters, a correlative use of complementary methods often yields a significant broader range of inform... Read more

3D computational anatomy of the scaphoid and its waist for use in fracture treatment
A detailed understanding of scaphoid anatomy helps anatomic fracture reduction and optimal screw position. Therefore, we analyzed the size and shape variations of the cartilage and osseous surface, the distribution of volumetric bone mineral density (vBMD), and if the vBMD values differ between a peripheral and a central screw pathway?
Forty-three fresh frozen hand specimens (17 females, 26 males) were analysed with high-resolution peripheral quantitative computed tomography (HR-pQCT) ... Read more

Mitochondria-rough-ER contacts in the liver regulate systemic lipid homeostasis
In this work, we studied mitochondria-rER contacts in vivo by serial section electron tomography (SSET) and 3D reconstruction analysis of cryo-fixed mouse tissue samples. We characterized this inter-organelle association as mitochondria tightly wrapped by sheets of curved rER (wrappER). Further, we used multi-omics and genetic approaches to obtain evidence that the wrappER is a distinct intracellular compartment and demonstrate the importance of wrappER-mitochondria contacts for v... Defect structure process maps for laser powder bed fusion additive manufacturing
Accurate detection, characterization, and prediction of defects has great potential for immediate impact in the production of fully-dense and defect free metal additive manufacturing (AM) builds. Accordingly, this paper presents Defect Structure Process Maps (DSPMs) as a means of quantifying the role of porosity as an exemplary defect structure in powder bed printed materials. Synchrotron-based micro-computed tomography (μSXCT) was used to demonstrate that metal AM defects follow predictable... Read more

Aging of a Pt/Al2O3 exhaust gas catalyst monitored by quasi in situ X-ray micro computed tomography
Catalyst aging effects were analyzed using X-ray absorption micro-computed tomography in combination with conventional characterization methods on various length scales ranging from nm to μm to gain insight into deactivation mechanisms.
For this purpose, a 4 wt% Pt/Al2O3 model exhaust gas catalyst was coated on a cordierite honeycomb and subjected to sequential thermal aging in static air at 950 °C for 4, 8, 12 and 24 hours. The ag... Read more

The paper proposes a new experimental methodology, based on ultrasonic measurements, that aims at evaluating the anisotropic damage in woven semi-crystalline polymer composites through new damage indicators. Due to their microstructure, woven composite materials are characterized by an anisotropic evolution of damage induced by different damage mechanisms occurring at the micro or mesoscopic scales. In this work, these damage modes in polyamide 6.6/6-woven glass fiber reinforced composites ha... Read more

The aim of this paper was to develop a model that can characterize the actual micropore structures in coal and gain an in-depth insight into water’s seepage rules in coal pores under different pressure gradients from a microscopic perspective. To achieve this goal, long-flame coals were first scanned by an X-ray 3D microscope; then, through a representative elementary volume (REV) analysis, the optimal side length was determined to be 60 μm; subsequently, by using Avizo software, the coal ... We describe in detail a methodology to estimate the effective elastic parameters of nodular cast iron, using micro-tomography in conjunction with multiscale finite elements. We discuss the adjustment of the image acquisition parameters, address the issue of the representative-volume choice, and present a brief discussion on image segmentation. In addition, the finite-element computational implementation developed to estimate the effective elastic parameters from segmented microstructural imag... Read more

Cyclostome bryozoans are an ancient group of marine colonial suspension-feeders comprising approximately 700 extant species. Previous morphological studies are mainly restricted to skeletal characters whereas data on soft tissues obtained by state-of-the-art methods are still lacking. In order to contribute to issues related to cyclostome ground pattern reconstruction, we analyzed the morphology of the neuromuscular system Cinctipora elegans by means of immunocytochemical staining,... 3D Reconstruction of Lipid Droplets in the Seed of Brassica napus
Rapeseed is one of the most important and widely cultured oilseed crops for food and nonfood purposes worldwide. Neutral lipids are stored in lipid droplets (LDs) as fuel for germination and subsequent seedling growth. Most of the LD detection in seeds was still in 2D levels, and some of the details might have been lost in previous studies. In the present work, the configuration of LDs in seeds was obtained by confocal imaging combined with 3D reconstruction technology in Brassica napus<... Read more
